چکیده

deposition and degradation. Local fibrin deposition and fibrinolysis are found at sites of wound healing and tissue remodeling. At these sites, the induction of apoptosis by fibrin degradation products could facilitate the clearance of damaged cells. Cross-linked fibrin is also a component of the matrix of most cancers,5 and degradation of fibrin by plasmin generated in the vicinity of tumor cells could support tumor cell apoptosis. The work of Guo et al, which focuses mainly on structural requirements of the proapoptotic molecule, not only raises many new questions related to the intracellular pathway–mediating apoptosis but also represents the basis for many new studies analyzing the physiologic relevance of apoptosis induction by fibrin degradation products. Lymphatic vessels and lymphangiogenesis in female cancer: mechanisms, clinical impact and possible implications for anti-lymphangiogenic therapies (Review).

Lymphatic vessels play a crucial role in a variety of human cancers, since invasion of lymphatic vessels by tumor cells and subsequent development of lymph node metastases significantly influences prognosis of cancer patients and therefore represent an integral part of tumor staging. Recent evidence on the important influence of lymphangiogenetic growth factors on intralymphatic cancer growth a...

A new target for simultaneous inhibition of hem- and lymphangiogenesis.

Toyono et al. established a new mediator of corneal hemand lymphangiogenesis: angiopoietin-like protein 2 (Angptl2). Angptl2 was expressed by the corneal epithelium and stroma after corneal suture injury, and increased infiltrating macrophages and expression of interleukin 1-b. RNAi-mediated knockdown of Angptl2 inhibited hemand lymphangiogenesis. Studies in conditional knock-in and knock-out m...

Prognostic value of lymphangiogenesis and lymphovascular invasion in invasive breast cancer.

OBJECTIVE The aim of this study was to investigate the prognostic relevance of lymphangiogenesis and lymphovascular invasion in a large cohort of breast cancer patients. INTRODUCTION Invasion of tumor cells into blood and lymphatic vessels is one of the critical steps for metastasis. The presence or absence of lymph node metastasis is one of the main decision criteria for further therapy. One...

Neuropilins: A New Target for Cancer Therapy

Recent investigations highlighted strong similarities between neural crest migration during embryogenesis and metastatic processes. Indeed, some families of axon guidance molecules were also reported to participate in cancer invasion: plexins/semaphorins/neuropilins, ephrins/Eph receptors, netrin/DCC/UNC5. Neuropilins (NRPs) are transmembrane non tyrosine-kinase glycoproteins first identified a...

Collaborative interplay between FGF-2 and VEGF-C promotes lymphangiogenesis and metastasis.

Interplay between various lymphangiogenic factors in promoting lymphangiogenesis and lymphatic metastasis remains poorly understood. Here we show that FGF-2 and VEGF-C, two lymphangiogenic factors, collaboratively promote angiogenesis and lymphangiogenesis in the tumor microenvironment, leading to widespread pulmonary and lymph-node metastases. Coimplantation of dual factors in the mouse cornea...

Prostaglandin E2 receptor EP4 as the common target on cancer cells and macrophages to abolish angiogenesis, lymphangiogenesis, metastasis, and stem-like cell functions

We previously established that COX-2 overexpression promotes breast cancer progression and metastasis. As long-term use of COX-2 inhibitors (COX-2i) can promote thrombo-embolic events, we tested an alternative target, prostaglandin E2 receptor EP4 subtype (EP4), downstream of COX-2. Here we used the highly metastatic syngeneic murine C3L5 breast cancer model to test the role of EP4-expressing m...
